CDX2 silencing downregulated Reg IV in gastric cancer cells. AGS (a, b) and MKN-45 (c, d) cells were transfected with the three CDX2 siRNAs for 48 h, and the CDX2 siRNA with the optimal silencing effect was chosen and used for subsequent experiments (left of a, c). CDX2 mRNA level was measured using real-time PCR, and the results were normalized to GAPDH (right of a, c). Moreover, western blotting was used to analyze Reg IV and CDX2 protein levels, β-actin was used as a loading control. Representative immunoblots are shown and the relative protein signal intensity was quantitatively analyzed using Image Lab software (b, d).
