Stable inhibition of HSP70 in AGS cell line by siRNA. (A) AGS cells were transfected with siRNA vector targeting HSP70. a. transient transfection; b. mono-clone cell; c. stable transfection. (B) The level of HSP70 mRNA was measured by real-time PCR. Left panel showed the representative images of PCR in AGS cells transfected with siRNA/HSP70 or empty vector. Quantitative results of real-time PCR were shown in the right panel. GAPDH was used as loading control. (C) The expression profile of HSP70 in transfected AGS cells was further confirmed by immunoblotting with β-actin as an internal control.
